This project will use Lifelines phenotypes, DEEP multi-omics, DEEP/DAG3 gut metagenomics, and host genetic data to investigate how lifestyle, socioeconomic, psychosocial, environmental, and medication-related exposures influence physiological and biochemical traits, molecular profiles, biological ageing, age-related diseases and mortality. We will identify exposure-associated proteomic, metabolomic and microbiome signatures; develop host- and microbiome-based ageing clocks and disease-risk models; and examine pathways linking exposures to ageing, disease and mortality. We will further assess molecular mediation, genetic susceptibility, host–microbiome interactions, and longitudinal trajectories to clarify disease mechanisms and identify potential biomarkers and intervention targets.
